Cultivation

In a series of experiments, the impact of Chlorella vulgaris on the growth of cucumber plants was investigated in hydroponics. This positive effect is attributed to the presence of soluble sugars and soluble amino acids (particularly glycine) in the algae infusion.

It was also found that the morphological characteristics of the cells, which were determined by the Goryaev camera, are similar to those of authentic Chlorella vulgaris. In addition, the ability of this strain to accumulate carotenes was determined by analysing the cell wall material for the presence of lutein, astaxanthin and fucoxanthin.

The results of the experiment showed that the optimum growth conditions for Chlorella sp. NTCU-2 were the combination of high light intensity and CO2 concentration. This resulted in a specific productivity value of 0.9 g dw l-1 in 5 days for Chlorella and 1 g dw l-1 in 10 days for Oedogonium. The results also demonstrated the efficient utilization of CO2 in this culture by maintaining the optimum oxygen tension.

Dyno(r)-Milling

The bead mill is a versatile tool that can be used to mechanically disrupt microalgae. To assess its effectiveness as a biorefinery tool, the recovery of proteins, carbohydrates and lipids from Chlorella vulgaris was tested using bead milling and enzymatic hydrolysis. The results demonstrate that bead milling is a suitable method to disrupt the cell wall of microalgae and improve lipid fractionation.

A DYNO(r)-Mill KDL A with a separation gap of 0.2 mm and a grinding chamber volume of 75 mL was used for the cell disruption experiments. The bead mill was filled to 65% with 0.4 mm Y2O3 stabilized ZrO2 beads and operated in batch recirculation mode at 25 degC.

The first experiment investigated the release of intracellular proteins during bead milling. As shown in Fig. 1, the yield of soluble proteins was approximately 10% after half a minute of bead milling. The high contact surface of the zirconium beads with the cells caused a rapid break down of the protein structures.

The second experiment evaluated the effect of bead milling and enzymatic treatments on the recovery of proteins, carbohydrates and lipids. The results demonstrate that bead milling significantly improved the protein and carbohydrate recovery compared to a control sample without bead milling. In addition, enzymatic treatment with lipase enhanced the recovery of lipids and carbohydrates.
